Probably serious with comical moments. The players influence the mood of the game, though, so it's also up to how they RP.

Who might be interested in a murder mystery RP? Probably Advanced and Adult.
It would be a classic whodunit tale from a previous decade, with "Clue" and mystery dinner parties as the inspiration....

My personal preference would be to lean towards a less "magical" style, so that vampires exist because of a disease (or something) and don't have inexplicable fantasy properties...
